SEARCHING FOR SOURCES OF WHEAT RESISTANCE TO INFECTION BY PUCCINIA STRIIFORMIS (YELLOW RUST) AS AN ELEMENT OF INTEGRATED PLANT PROTECTION

The aim of the project is to search for sources of wheat resistance to infection by Puccinia striiformis. SNP markers ware used for this purpose. The 500 wheat lines differing in disease resistance.

The task of the project "Development of a large number of polymorphic SNP markers covering the whole wheat genome, and the identification of markers linkage with the resistance feature" was carried out at the Department of Biochemistry and Biotechnology by dr hab. Łukasz Wolko, e-mail lukwolko@gmail.com.. The marker-assisted selection method (MAS) is an important tool for increasing the efficiency of breeding procedures. The method consists in the early selection of a cross-breeding material, based on molecular markers linkaged to regions of the genome responsible for the expression of the trait (e.g. resistance). In the case of crop plants, it is necessary to study at least several of thousands markers to develop the MAS. Recently the methods based on new generation sequencing technologies (NGS - next generation sequencing) provide new efficient sources for SNP type markers. One of the methods using NGS in genotyping is SBG (sequence-based genotyping). This method, based on the reduction of genomic complications, and is very efficient, simple and relatively cheap. Thanks to the highly efficient, parallel sequencing of short fragments derived from different genotypes and bioinformatics comparative analysis, many thousands of SNP markers are obtained in a short time. These markers can be directly used for mapping quantitative or qualitative traits, and in further works, for marker assisted selection.
